"Differentiation-dependent Human Papilloma Virus (HPV) amplification depends on the activation of the ataxia telangiectasia mutated (ATM) DNA damage response. The structural maintenance of chromosomes (SMC) family of proteins are part of the cohesin and condensin complex, but also regulate homologous repair, chromatin looping and organization, and are necessary for ATM signaling. SMC-1, a cohesin protein, is a critical component of the ATM DNA damage response and can directly interact with DNA alongside the DNA insulator and chromatin looper CCCTC-binding factor (CTCF). Our studies demonstrate a role for both SMC-1 and CTCF in the HPV-31 differentiation-dependent lifecycle in both patient-derived and stably transfected HPV positive cell lines. Ongoing studies will investigate the importance of CTCF-mediated DNA looping interactions in the HPV lifecycle and mechanisms by which the virus leverages the ATM DNA damage response towards its productive lifecycle. "
