"We have developed approaches to single particle track HCV in polarized three dimensional liver organoids to define the complex pathway of its endocytosis. Additionally, we developed dual RNA in situ hybridization detection of HCV (+) and (-) RNA to characterize the spatiotemporal dynamics of HCV replication. This revealed, among other things, a previously unappreciated biphasic replication strategy wherein a low level of transient replication occurs early in infection to create reserve genome copies. This is followed by the formation of dedicated replication compartments and robust replication."
