"Advances in Single-cell Combinatorial Indexing in Neuroscience and Cancer"

Abstract:

Single-cell genomics technologies have become nearly ubiquitous across a wide range of life science disciplines due to the ability to deconstruct the molecular profiles of complex tissues or systems into its cellular components. However, most studies deploy single-cell techniques based on assay availability and throughput, limiting the scope and scale of what can be investigated. In the Adey Lab we have developed a variety of technologies to address the breadth of properties that can be profiled as well as the scale to enable larger, and more powered, studies. In recent advances we have developed a robust and high-throughput assay to assess DNA methylation, a notoriously difficult property, and applied it to the human brain. We have also developed a highly-accessible strategy for high-throughput single-cell genome sequencing, producing up to tens-of-thousands of cell profiles using widely available off-the-shelf reagents, which we have begun to deploy in several areas of cancer research. These techniques also form the basis of novel multi-omic profiling tools that enable multiple properties to be assayed simultaneously from the same cells.
